Description
TheR-Spondin(Rspo)proteinsbelongtotheRspofamilyofWntmodulators.Currently,thefamilyconsistsoffourstructurally-related,secretedligands(Rspo1–4),allcontainingfurin-likeandthrombospondinstructuraldomains.TheRspoproteinscaninteractwiththeFrizzled/LRP6receptorcomplexinamannerthatcausesthestABIlization,andresultingaccumulation,oftheintracellularsignalingprotein,β-catenin.Thisactivityeffectivelyactivatesandincreasesthesubsequentnuclearsignalingofβ-catenin.R-SpondincanalsobindtothepreviouslydiscoveredG-proteincoupledreceptors,LGR-4 andLGR-5. Rspo/β-cateninsignalingcanactasaninducerofthetransformedphenotype,andcanalsoregulatetheproliferationanddifferentiationofcertainstemcellpopulations.RecombinantHumanR-Spondin-2 isa24.4 kDaproteinconsistingof212 aminoacidresidues.Duetoglycosylation,R-Spondin-2 migratesatanapparentmolecularweightofapproximately30.0 kDabySDSPAGEanalysisunderreducingconditions.
